La 640 est la première Ferrari entièrement conçu par John Barnard. La nouvelle réglementation impose des moteurs de 3,5 litres et précise que les pédales ne doivent pas dépasser l'axe des roues avant. La carrosserie de la monoplace est très étroite à l'arrière et les entrées d'air des pontons sont très avancées. Lombardi à dessiné un moteur V12 à 65° (600 ch) accouplé à une boite semi-automatique dont la première réalisation date de 1981.
Mansell dispute avec Berger les seize Grands Prix sous la direction de course de Cesare Fiorio. Mansell triomphe dans le premier grand Prix (Brésil) avant une succession d'abandons. Il est de nouveau premier en Hongrie, deuxième en France et en Grande-Bretagne. Berger, souvent hors piste, est cependant premier au Portugal, deuxième en Italie et en Espagne.
Mansell est quatrième au championnat pilotes et Ferrari troisième des constructeurs.
